Effects of TGF-β on Growth and Invasion of Human Oral Squamous Cell Carcinoma Cell Lines

Although TGF-β is known to be related tumor proliferation, invasion and epithelial-mesenchymal transition (EMT), its effects on oral carcinomas remain unclear. This study aimed elucidate the of squamous cell carcinoma (OSCC). In present in vitro study, three human OSCC lines; SAS (tongue cancer), Ca9-22 (mandibular gingival cancer) HSC-3 (cervical lymph nodes metastasis tongue were treated with TGF-β1 or signaling inhibitors, assayed for proliferation invasion. Expressions EMT-related markers; E-Cadherin, N-Cadherin, Snail Slug, as well matrix metalloproteinases (MMPs) evaluated by immunofluorescence staining RT-qPCR. treatment induced morphological change spindle shape, significant (p<0.05), increase capacity all lines (p<0.05). Inhibition significantly suppressed By TGF-β1, reduced expression E-Cadherin enhanced expressions Slug observed (poorly differentiated OSCC), whereas was (well OSCC) variable depending concentration (lymph node metastatic OSCC). MMPs lines. increases lines, while inhibition suppresses proliferation. may induce EMT enhancing transcription activity, suppressing epithelial markers augmenting mesenchymal poorly OSCC, but both mesenchymal-epithelial (MET) OSCC.
